Abstract
To investigate the mutual dependence of calcitonin gene-related peptid e (CGRP) and acetylcholine release, we examined the effect of a cholin esterase inhibitor neostigmine on the release of CGRP-like immunoreact ivity in rat phrenic nerve-hemidiaphragm muscle preparation, and conve rsely, the effect of CGRP on [H-3]acetylcholine release from motor ner ve terminals loaded with [H-3]choline in the same preparations of mice . Release of CGRP-like immunoreactivity was increased by electrical ne rve stimulation (train of 40 pulses of 200 mu s pulse duration and fre quency of 50 Hz applied every 10 s) in the whole preparation but not i n the segmental preparation containing the endplate region. Neostigmin e (0.1-0.3 mu M) enhanced the resting release of CGRP-like immunoreact ivity in a concentration-dependent manner, whereas it depressed the ne rve-evoked release of CGRP-like immunoreactivity. CGRP (1 mu M) added to perfusate decreased nerve-evoked [H-3]acetylcholine release. These results suggest that CGRP, which is released by electrical nerve stimu lation or a cholinesterase inhibitor in intact skeletal muscles, negat ively modulates nerve-evoked acetylcholine release.
