Abstract
Although pituitary adenylate cyclase activating polypeptide (PACAP) has bee n shown to increase cardiac force of contraction and to change the heart ra te, the effect of PACAP on cyclic (c) AMP production in the atrium still ha s to be defined. In the present experiments, a simple protocol was develope d for the evaluation of cAMP production in real-time base in the perfused b eating left atria. The PACAP27-induced cAMP efflux in the atrial perfusate reflected changes in the production of cAMP in the atrial tissue. cAMP effl ux was measured as an indicator of cAMP production in beating perfused rabb it atria. PACAP27 increased cAMP production in a dose- and time-dependent m anner with a minor effect on atrial dynamics. These results suggest that PA CAP27 has other roles besides control of force of contraction through cAMP production in the atrium. (C) 2000 Elsevier Science B.V. All rights reserve d.
