Abstract
beta-amyloid (A beta) is toxic to cells and has been identified as the majo r participant in the pathogenesis of Alzheimer's disease. In both the corpu s luteum organ culture model of apoptosis and in neuronal cells, A beta was able to mimic the action of superoxide dismutase and prevent apoptotic end onucleolytic DNA fragmentation at low concentrations. Furthermore the longe r form of A beta (A beta(1-42)) was more potent than A beta(1-40) in reduci ng apoptosis. At increasing A beta concentrations a loss of anti-apoptotic activity became evident. In addition, the anti-apoptotic effect of A beta w as dependent upon copper binding to the peptide. Since copper is a transiti on metal and is integral to the activity of the superoxide dismutase enzyme , it is suggested that the novel anti-apoptotic effect of A beta may be med iated by superoxide dismutase-like activity.
