Abstract
The modulatory effect of PACAP(27) on NMDA receptor channel activity i n cultured chick cortical neurons was investigated using the outside-o ut recording mode of the patch clamp technique. Channel opening freque ncy elicited by 20 mu M NMDA, or 20 mu M NMDA plus 1 mu M glycine, was potentiated in the presence of 100 nM PACAP(27) and inhibited with 10 00 nM PACAP(27). These effects were reversible on washout and reduced when glycine concentration was increased to 10 mu M, but were not affe cted by the PACAP antagonist PACAP(6-27) (1 mu M) or the GTP inhibitor GDP-P-S (100 mu M). It is suggested that PACAP(27) may exert its modu latory action on NMDA receptor channel activity through the glycine si te(s). (C) 1998 Elsevier Science B.V.
