Abstract
H-1 AND P-31 NUCLEAR magnetic resonance (NMR) spectroscopy were used t o study the role of external pH (pH(o)) in regulation of intracellular pH (pH(i)) in the cerebral cortex in vitro. Lowering the pH of the ox ygenated superfusion medium (pH(b)) from 7.5 to 7.0-6.8 decreased pH(i ) from 7.33 +/- 0.08 to similar to 7.00 independently of the presence of HCO3- and medium buffering power. Reduction of medium (pH(b) 6.8) b uffering power during hypoxia severely acidified pH(i) independently o f the presence of HCO3-. Following hypoxia, recovery of pH(i) was not sensitive to pH(b) provided that medium buffering was maintained. Lact ate removal proceeded independently of pH(b), HCO3- and external buffe ring power. These results indicate that decrease in extracellular buff ering capacity has a feed-back effect on pH(i), in the cerebral cortex .
